- Immunophenotyping is the identification, classification, and quantification of individual cells in a heterogenous population based on the types of markers or antigens present on their surface, nucleus, or cytoplasm. - Cell proliferation assays use cell-permeable fluorescent dyes to covalently label cells of interest prior to stimulation with an antigen, peptide, mitogen, allogeneic cells, or other test articles. Cell proliferation is then quantitatively tracked by measuring the dilution of the fluorescent dye due to cell divisions over time. Our team has expertise in developing and qualifying CD4+ and CD8+ T cell proliferation assays for virology and immune-oncology applications.
- Cytotoxicity assays quantify the efficacy and kinetics of immune effector cells in eliminating target cells. Unlike classic approaches using radio-actively labelled target cells, we employ the cell permeable calcein-AM dye enabling high-throughput flow cytometry analysis. Our team has developed custom cytotoxicity assays for a variety of effector cell types, including both wild-type and genetically engineered T cells and NK cells.
- Intracellular cytokine staining (ICS) is an approach for measuring levels of important cytokines within individual cells. After stimulation or treatment of a cell population of interest to induce cytokine production, cells are fixed and permeabilized to enable intracellular staining of cytokines of interest with fluorescently labelled antibodies. We employ ICS as both a standalone technique and paired with other functional immunology assays to provide deeper insights.
